I have just finisjed my deathstrike missile and am itching to unleash it upon my foe... However, I feel I need some adivce on building my army to compliment such an awesome weapon.
I am most likely to play 1500pts, and I normally play against either space marines or crons...
I guess the most important thing is keeping the missile alive unitl it is ready to fire, and then choosing my targets carefully...
Just a quick thought, but an army containing plenty of mortars and snipers that could sit back around the missile could work well. Cleverly use the pinning to clump units together and your missile will have a tasty target to go for.
You'd want something to screen/protect it, so either a russ squadron or (more likely) another artillery unit.
You'll then likely want to be playing a gunline, but a mobile-ish one so that you can stay WELL away from your target for the DSM. Some vets in chims maybe, a platoon or 2 with a large conscript squad and commissar would give a nice tarpit to bog down a nasty. Some mortar squads would help with pinning down your opposition, possiblty some ratlings too and a PBS.

Just stick it in a corner and give your opponent more direct threats to worry about, like meltas, flamers, and Demolisher cannons.

not really that effective of a weapon in a 1500 game, maybe if its a big battle its useful, but if you want something with a large blast radius, use a manticore, you can even use it multiple times

In addition to providing a little protection for your Deathstrike (a cover save would suffice), have some good CC units in your list. Either you opponent will focus on the Deathstrike and let the rest of your army have fun (the real reason to take a Deathstrike, in my opinion) or he'll spread out to minimize its effect--and what better way to spread out than a headlong charge into your lines so you end up nuking your own men, too? The nuke means he can't sit back. CC units like Rough Riders or Straken mean he can't charge in. And everything else means medium range isn't fun, either. Watch your opponent try to solve that dilemma!

I'd take a big ooutflanking platoon with Al'rahem, or soething similar. Just to keep them in their deployment zone while the gunline blasts their army away.
