I have lots of questions:
Big Shootas, Burnas and Rokkit Launchas, which one of these is the most balanced vs multiple armies?
Which one is the most cost effective?
The best in a KOS army?
The best against (insert race here).
What should you give them to/ mount them on (Vehicles, Troops, etc.)?
These weapons all seem very powerful in their own right, but what do they work best against?
What would you take in a tournament?
Are there certain weapon that are better in some clans?
Sorry if my questions seem somewhat scattered. I just can't pick which one, they're all so good.

My personal fave gun is the Big Shoota. An assault bolter is good in anyones language. Because of the 3 shots it fires every turn, it gives me a better chance to overcome the orks BS. All backed up by a long range. Tasty
I must admit I am also a big fan of the rokkit launcha with ammo runts too though, in fact my tankbusta's with four rokkit launcha is my favourite unit.

In my opinion, I believe that Big Shootas are the superior and more cost effective weapons when confronting an unknown foe, because Rokkit Launchas have only one shot and are 24" range and Burnas are just too short ranged. Big Shootas are Assault 3 and have a Strength of 5 which isn't too bad and like chemicalcaveman said, it compensates for the Orks' poor Ballistic Skill. Big Shootas are best on standard boyz, especially those that will hang back (if any). They are best against light troops such as Eldar Guardians (anything with a 5+ or worse armour save).
However, you should mount Rokkit Launchas on anything that is twin-linked (Wartraks for example) as to give you some tankbusting power whilst maintaining accuracy. I suppose Rokkit Launchas would be good against Space Marines, their evil counterparts and any other army that predominantly has 3+ or 4+ armour saves. Then, of course, Rokkit Launchas are the main anti-tank weapon for the Orks.
Burnas are good for those who will definitely see combat, as its ability to be used as a power weapon or alternatively as a flamer comes in useful - the flamer should be good for clearing out lightly-armoured troops whilst the power weapon would be good against heavily armoured troops. Burnas would probably be good against swarms such as Tyranids but would serve adequately against heavily armoured troops.
I don't think that clans benefit from using particular weapons - the clans are all mainly fluff unless you are taking a special character like Ghazghkull Mag Uruk Thraka. I hope this has been helpful.
As was noted before, it really depends on which foe you're facing...
My skarboyz always get a burna (strength 4 powerweapon? Why THANK you!) and I usually have a warbuggy or two with a TL-RL. I have a warboss and a nob mounted on a boar, both with burnas, to take care of anything else that happen to vex me (snakebite rules, wee!).
As for what to take in a KoS list... I've seen one list that takes two seperate Tankbustas, one armed with rokkit launchas, the other with big shootas. Mount them in a trukk and you're set, and be sure to add a BoBS to the big shoota trukk. The Tankbustas give the shootas an effective strength of 6, which lets you take out quite a bit. Give the units an armoured top and you're good to go.
Expensive as all hell, and probably really not all that useful, but you'd be amazed with the carnage they can pull out.
